Lab & Project Overview
The laboratory has established patient-derived 3D intestinal organoid (colonoid) models for inflammatory bowel disease (IBD), including ulcerative colitis. The current project centres on developing autologous organoid co-culture systems incorporating Lamina Propria Mononuclear Cells (LPMCs) and Fibroblasts.
Key Responsibilities & Technical Competencies
Organoid & Cell Culture:
- Adult stem cell-derived human intestinal organoid culture, maintenance and optimization.
- Primary cell culture: fibroblast and lamina propria mononuclear cell (LPMC) isolation and characterization.
- Co-culture model development and optimization (organoid + primary cell integration).
Characterization & Assessment:
- Flow cytometry (FACS) - cell sorting, gating, population analysis.
- Immunofluorescence and immunocytochemistry (ICC) staining.
- Live-cell imaging and confocal microscopy.
- Functional cell-based assays.
Molecular & Genomic Analysis:
- RT-qPCR and Western blotting / multiplex protein assay.
- scRNA-seq sample preparation and library prep.
- Transcriptomics data interpretation.
